Effect of papain on musculo-skeletal growth of rats

Abstract
Crude papain was injected for 28 days into 12 mature rats and this hindered growth of long bones. Thirty nine male ature rats were used in this experiment, three killed at the start and the remainder were randomly allocated into three groups. Two types of controls were used with the treated group as a comparison: littermates were pairfed with those receiving papain and other littermates were fed ad libitum throughout the experiment. All rats were killed at the end of the period of the injection and X-rays were taken at the start and the end of the experiment. The tibia, femur and humerus of the papain-treated rats were short (P<0.001) compared to those of pairfed and control animals but there was no differential effect on diameter of these bones. Mean bones length for control, treated and pairfed animals were 39.21, 37.76, and 38.86 for tibia respectively and 34.17, 32.27 and 33.14 for femur and 28.14, 26.75 and 27.55 for the humerus respectively. There was no papain treatment effect on muscles attached to these bones.There was no treatment effect between the papain-treated animals and the pairfed in body weight gain and body length but there was a significant difference (P<0.001) in tail length between control, treated and pairfed (their means were 5.6, -4.6 and 0.8 respectively). Crude papain affects the epiphyseal cartilage of long bones thus hindering growth of bones.
